I was wondering whether the Tufts and Emory Early Decision II rounds on January 1st help your chances of admission.

The Tufts EDII does slightly help your chances of admission - it is treated the same as ED I in terms of admission. You are not subject to the problems seen at the end of the pile - i.e. you are not going to be rejected because you are a potential Spanish major and they have too many potential Spanish majors. It does slightly help your chances, but the improvement is not huge. No idea about Emory.

If you get rejected in Tufts ED I can you try again in ED II?

I think if you are deferred in round one, then you are considered along with RD candidates.
